美国Plant Cell Technology公司为商业及研究用植物组织培养实验室和植物种植者开发、生产了旗舰产品PPM™ (Plant Preservative Mixture),它是一种广谱抗微生物剂,可以杀死细菌和真菌细胞,防止真菌孢子的萌发,并在较高浓度下能消除内源性污染的外植体。研究表明PPM™活性成分可以渗透过真菌或细菌细胞壁,抑制柠檬酸循环和电子传递链等中心代谢循环中的关键酶的活性,并且可以抑制培养基中的单糖和氨基酸向真菌和细菌细胞的运输。PPM™能有效地抑制植物细胞和植物组织培养中的通过空气、水、人传播的微生物污染及内源性污染,而不会影响体外种子发芽、愈伤组织增殖和愈伤组织的再生。并且,PPM™具有热稳定性,可以和培养基一起灭菌,同时比常用抗生素价更低廉,可以作为植物组织培养基的标准成分。

 

The principal PCT scientist involved in the development of the PPM™ application is Dr. Assaf Guri. Dr. Assaf Guri holds degrees in genetics, applied genetics and plant breeding from the Hebrew University in Jerusalem and Michigan State University in the US. Before joining Plant Cell Technology, Inc., Dr. Guri worked with the Volcani Agricultural Research Center in Israel, Michigan State University in East Lansing, Michigan and DNAP in New Jersey.

The UC Davis/NIH NeuroMab Facility is a national monoclonal antibody-generating resource funded by the National Institutes of Health (NIH). The mission of the UC Davis/NIH NeuroMab Facility is to provide a unique neuroscience-based approach to generating mouse monoclonal antibodies optimized for use in mammalian brain (NeuroMabs). NeuroMabs are generated from mice immunized with synthetic and recombinant immunogens corresponding to components of the neuronal proteome as predicted from genomic and other large-scale cloning efforts. Comprehensive biochemical and immunohistochemical analyses of human, primate and non-primate mammalian brain are incorporated into the UC Davis/NIH NeuroMab Facility screening procedure. This yields a subset of mouse mAbs that are optimized for use in brain (i.e., NeuroMabs): for immunohistochemical- based imaging studies of protein localization in adult, developing and pathological brain samples, for biochemical analyses of subunit composition and post-translational modifications of native brain proteins, and for proteomic analyses of native brain protein networks.

艾乐美生物科技有限公司(AllerMAbs Co., Ltd.)成立于2015年,是台湾一家专注于生产高亲和力、高纯度并具有过敏原特异性的嵌合人类IgE单克隆抗体的生物公司。公司创办人团队在科研过程中发现市面上缺乏多样的IgE单克隆抗体,特別是具有过敏原特异性的人IgE 单抗。如果能制备出这种抗体,将有助于IgE 相关领域的研究工作。因此,AllerMAbs公司团队开始思考与研究利用基因克隆动物来制造人类 IgE 抗体的可行性,经过数年的努力于 2014年开发出人类IgE 抗体基因嵌入小鼠,通过杂交瘤技术 (Hybridoma technology),成功地研制出分泌嵌合人类IgE 单抗的 IgE 杂交瘤与纯化 IgE 抗体。经测试,此种嵌合人类IgE 单抗的物化性质与对免疫细胞的活性,皆与人类IgE 抗体一致,研究成果于2015 年发表在国际学术期刊上。

 

AllerMAbs的产品解决了目前人类IgE抗体不易制备的问题,让嵌合人类IgE抗体可以成为人类IgE抗体的替代品,并提供一个更稳定、更符合血清IgE检测系统的校正品,让临床血清IgE的检测结果更加准确与可靠。

 

其利用动物基因转殖技术,将组成人类IgE抗体分子的ε重链(Heavy chain)和κ轻链(Light chain)的编码基因(Encoding genes),分別嵌入(Knock-in)小鼠胚胎干细胞(Embryonic stem cells, ES cells)的γ1重链和κ轻链基因的位置,接着把胚胎干细胞分別植入孕母鼠体内,并将其带有嵌入基因之成鼠交互配种,最后得到带有人类IgE抗体重链和轻链基因的基因嵌入小鼠(HεκKI mice),且体内能制造完整的嵌合人类IgE抗体分子。

The UC Davis/NIH NeuroMab Facility is a national monoclonal antibody-generating resource funded by the National Institutes of Health (NIH). The mission of the UC Davis/NIH NeuroMab Facility is to provide a unique neuroscience-based approach to generating mouse monoclonal antibodies optimized for use in mammalian brain (NeuroMabs). NeuroMabs are generated from mice immunized with synthetic and recombinant immunogens corresponding to components of the neuronal proteome as predicted from genomic and other large-scale cloning efforts. Comprehensive biochemical and immunohistochemical analyses of human, primate and non-primate mammalian brain are incorporated into the UC Davis/NIH NeuroMab Facility screening procedure. This yields a subset of mouse mAbs that are optimized for use in brain (i.e., NeuroMabs): for immunohistochemical- based imaging studies of protein localization in adult, developing and pathological brain samples, for biochemical analyses of subunit composition and post-translational modifications of native brain proteins, and for proteomic analyses of native brain protein networks.
